The significance of the GPLD1-TNAP pathway is underscored by striking animal experiments. Young mice engineered to overproduce TNAP developed memory and cognitive problems resembling those of elderly animals. Conversely, when researchers reduced TNAP levels in mice equivalent in age to 70-year-old humans, the blood-brain barrier tightened, inflammation fell, and memory performance improved — suggesting it is never too late to benefit from intervention.

Led by Dr. Saul Villeda of the UCSF Bakar Aging Research Institute, the findings open a new avenue for fighting neurodegenerative diseases like Alzheimer's. Scientists believe drugs that mimic GPLD1's protein-trimming action could one day offer protection even for people unable to exercise regularly.
